Medarot Club
- Koishimaru Tensan - The Protagonist. His initial Medarot is Chrotojil or Shinzan depending on the version.
- Saki Oomura - One of Koishimaru's friends and possible romantic interest, she hates losing. Her primary Medarot is Piccoli.
- Yamato Shijimi - He founded the Medarot Club with Hikoo. His main Medarot is Hashimukou.
- Hikoo Tamayasu - Koishimaru's rival and the son of the mayor of Susutake Village. His main Medarot is the opposite version of Koishimaru's.
- Asahi Hyoumon - He is a bad boy who always hangs around with Osamu, and is the leader of the gang. His main Medarot is City Rusher.
- Osamu Samehada - A bad boy who always hangs out with Asahi. His main Medarot is Snow Fenrir.

- Konoha Tateha - She is a library committee member and a childhood friend of Asahi and Osamu. She is one of the romantic options. Her main Medarot is Pink Rabbie.
- Beni Shijimi - Yamato's younger sister. She adores Koishimaru and is one of the romantic options. Her main Medarot is Sanizurau.
- Kaoru Tsuyukusa - She is the daughter of a boat shop owner in Susutake Lake. She is one of the romantic options, but exclusive if played on Game Boy Advance. Her main Medarot is Gairain.
- Tsuguo Nino - He has a complex about always being second best at whatever he does. His main Medarot is Mauritius.
- Gretel - An Austrian Medarotter, she is Hansel's twin sister. Her main Medarot is Kimishadow.
Susutake Village
- Densuke Tamayasu - Hikoo's father. He is the mayor of Susutake Village and comes up with various plans to revitalize the village , but they rarely work. His main Medarot is Rockin' Kan.
- Kageo Sorifugi - Deputy mayor of Susutake Village. Despite holding the important position, he lives with his wife in a small apartment. His main Medarot is Magastron.
- Ito Arakune - The game's main antagonist. She is the village chief's secretary. Her main Medarot is Hardness Ten.
- Akane Aki - He is Koishimaru's teacher. He also happens to be the advisor for the Medarot Club. His main Medarot is Macularius.
- Gunbai Miyama - He is a part-time teacher at Susutake Elementary School and served as the school advisor until Akane became the advisor. His main Medarot is Eryops.
- Hamaki Usmon - He is a physical education teacher who is the advisor for the archery club and has a strong physique. His main Medarot is El Head.
- Michio Hanmyouji - A taxi driver who calls himself " the taxi driver of love and justice ", he gets to know Koishimaru when he takes him to school when he is about to be late on his first day at the new school. His main Medarot is Road Lord.
- Mr. Kabashira - The new referee for the robattles in Medarot 5. He seems to still be a rookie.
- Yakumo Arakune - She is the older sister of Ito, the manager of Seidou Academy. Her main Medarot is Inaosen.
- Takumi - A researcher at the Medarot Research Institute.
